What I always do when I have to play a song in drop D is I pluck the 6th and 4th strings simultaneously while keeping my left hand on the tuning peg for the 6th!. Keep plucking them and tuning your 6th down until they sound very simmilar, you'll know when you hit the sweet spot!. When you want to tune back up, repeat the process using the 1st instead of the 4th!.
